Bacterioferritin of magnetospirillum gryphiswaldense is a heterotetraeicosameric complex composed of functionally distinct subunits but is not involved in magnetite biomineralization

In particular, the existence, nature, and location of possible mineral precursors of magnetite are not clear. One possible precursor has been suggested to be ferritin-bound ferrihydrite. To clarify its role for magnetite biomineralization, we analyzed and characterized ferritin-like proteins from the magnetotactic alphaproteobacterium Magnetospirillum gryphiswaldense MSR-1, employing genetic, biochemical, and spectroscopic techniques. Transmission M{\"o}ssbauer spectroscopy of the wild type (WT) and a bacterioferritin (bfr) deletion strain uncovered that the presence of ferrihydrite in cells is coupled to the presence of Bfr. However, bfr and dps deletion mutants, encoding another ferritin-like protein, or even mutants with their codeletion had no impact on magnetite formation in MSR-1. Thus, ferritin-like proteins are not involved in magnetite biomineralization and Bfr-bound ferrihydrite is not a precursor of magnetite biosynthesis. Using transmission electron microscopy and bacterial two-hybrid and electrophoretic methods, we also show that MSR-1 Bfr is an atypical representative of the Bfr subfamily, as it forms tetraeicosameric complexes from two distinct subunits. Furthermore, our analyses revealed that these subunits are functionally divergent, with Bfr1 harboring a ferroxidase activity while only Bfr2 contributes to heme binding. Because of this functional differentiation and the poor formation of homooligomeric Bfr1 complexes, only heterooligomeric Bfr protects cells from oxidative stress in vivo. In summary, our results not only provide novel insights into the biomineralization of magnetite but also reveal the unique properties of so-far-uncharacterized heterooligomeric bacterioferritins. IMPORTANCE Magnetotactic bacteria like Magnetospirillum gryphiswaldense are able to orient along magnetic field lines due to the intracellular formation of magnetite nanoparticles. Biomineralization of magnetite has been suggested to require a yet-unknown ferritin-like ferrihydrite component. Here, we report the identification of a bacterioferritin as the source of ferrihydrite in M. gryphiswaldense and show that, contrary to previous reports, bacterioferritin is not involved in magnetite biomineralization but required for oxidative stress resistance. Additionally, we show that bacterioferritin of M. gryphiswaldense is an unusual member of the bacterioferritin subfamily as it is composed of two functionally distinct subunits. Precise anatomical dissection of the human temporal bone is essential and is fundamental for middle ear surgery. We assessed the possible application of a virtual reality temporal bone surgery simulator to the education of ear surgeons. Seventeen ENT physicians with different levels of surgical training and 20 medical students performed an antrotomy with a computer-based virtual temporal bone surgery simulator. The ease, accuracy and timing of the simulated temporal bone surgery were assessed using the automatic assessment software provided by the simulator device and additionally with a modified Final Product Analysis Scale. Trained ENT surgeons, physicians without temporal bone surgical training and medical students were all able to perform the antrotomy. However, the highly trained ENT surgeons were able to complete the surgery in approximately half the time, with better handling and accuracy as assessed by the significant reduction in injury to important middle ear structures. Trained ENT surgeons achieved significantly higher scores using both dissection analysis methods. Surprisingly, there were no significant differences in the results between medical students and physicians without experience in ear surgery. The virtual temporal bone training system can stratify users of known levels of experience. This system can be used not only to improve the surgical skills of trained ENT surgeons for more successful and injury-free surgeries, but also to train inexperienced physicians/medical students in developing their surgical skills for the ear.",author = "Robert Linke and A. Atypical iron storage in marine brown algae: a multidisciplinary study of iron transport and storage in Ectocarpus siliculosus

The iron uptake and storage systems of terrestrial/higher plants are now reasonably well understood, with two basic strategies for iron uptake being distinguished: strategy I plants use a mechanism involving induction of Fe(III)-chelate reductase (ferrireductase) and Fe(II) transporter proteins, while strategy II plants utilize high-affinity, iron-specific, binding compounds called phytosiderophores. In contrast, little is known about the corresponding systems in marine, plant-like lineages, particularly those of multicellular algae (sea- weeds). Herein the first study of the iron uptake and storage mechanisms in the brown alga Ectocarpus siliculosus is reported. Genomic data suggest that Ectocarpus may use a strategy I approach. Short-term radio-iron uptake studies verified that iron is taken up by Ectocarpus in a time- and concentration-dependent manner consistent with an active transport process. Upon long-term exposure to 57Fe, two metabolites have been identified using a combination of M{\"o}ssbauer and X-ray absorption spectroscopies. These include an iron–sulphur cluster accounting for \textasciitilde{}26\% of the total intracellular iron pool and a second component with spectra typical of a polymeric (Fe3+O6) system with param- eters similar to the amorphous phosphorus-rich mineral core of bacterial and plant ferritins. Fachkräftemangel in Deutschland: Umfrage zu Erwartungen von Chefärzten an junge Mitarbeiter

Die Qualit{\"a}t der Ausbildung scheint der wichtigste Faktor f{\"u}r die Wahl einer Klinik zu sein. Gleichzeitig nimmt die Eignung der Bewerber ab. F{\"u}r die Hals-, Nasen- und Ohrenheilkunde (HNO) liegen zur Eignung und zuk{\"u}nftigen Bewerberlage keine Ergebnisse vor. Methode: Es wurde ein Fragebogen nach anerkannten Richtlinien entwickelt und an 160 Fachabteilungen f{\"u}r HNO per E-Mail versendet. Gefragt wurde nach Gr{\"o}{\ss}e und Ort der Abteilung, Anzahl geeigneter bzw. ungeeigneter Bewerbungen, ferner nach heutiger und zuk{\"u}nftiger Verf{\"u}gbarkeit von Mitarbeitern sowie den Anforderungen aus Sicht des Chefarztes. Ergebnisse: Die R{\"u}cklaufquote lag bei 34\% (54 Frageb{\"o}gen). Die Kliniken erhalten im Durchschnitt 20 Bewerbungen pro Jahr, wobei 36\% der Bewerber ungeeignet sind. In den neuen Bundesl{\"a}ndern bewerben sich mehr {\"A}rzte als in den alten – ohne Unterschied bei der Anzahl ungeeigneter Bewerber. Universit{\"a}tskliniken erhalten signifikant mehr Bewerbungen als andere Kliniken. Mit der Gr{\"o}{\ss}e der Abteilung steigt die Anzahl und Qualit{\"a}t der Bewerber. Fast 60\% der Befragten erwarten zuk{\"u}nftig eine schlechter werdende Bewerberlage, fast 40\% der Befragten aus Kliniken mit mehr als 50 Betten eine bessere oder gleichbleibende Situation. Chef{\"a}rzte erwarten als wichtigste Voraussetzung f{\"u}r Mitarbeiter Engagement, eigenst{\"a}ndiges Lernen und Teamf{\"a}higkeit. Schlussfolgerungen: Die Breite und Verbindlichkeit der Ausbildung sowie strukturierte Ausbildungsprogramme sind ein wichtiger Wettbewerbsfaktor f{\"u}r die Rekrutierung junger Mitarbeiter. Hier haben gro{\ss}e Abteilungen und Universit{\"a}tsklinika Vorteile. Based on inhibitor studies it appears that iron is taken up directly as Fe(iii). However if a reductive step is involved the Fe(ii) must not be accessible to the external environment. Because these tetramic acids also appear to bind iron with appreciable affinity it was suggested that metal binding might contribute to their biological activity. Discussion about the effects of Mössbauer source line broadening

Computer calculations of emission line broadening of M{\"o}ssbauer sources with various effective thicknesses using the quantum mechanical theory of the emission spectrum proposed by Odeurs and Hoy, Nucl. Instrum. Methods Phys. Res. B, 254 (2007) 143-148 [1] are made. Corresponding calculations are performed using two simpler classical models giving an account of the non-resonant absorption of the radiation. Differences between the results obtained with the theory of Odeurs and Hoy, other model calculations and experimental data, and also inaccuracies, which could give rise to unusual large broadenings, are discussed. Two experimental tests are described, which unambiguously show that the broadenings calculated in [1] as a result of source line self-absorption broadening (age broadening) are huge and unrealistic. These quantitative evaluations of the age broadening yields an increase of line width of about 0.0004 mm/s for the decrease of 57Co/57Fe source activity by 1 mCi. Our conclusion is that the predicted observation of {"}hole burning{"} line profile could be possible, but only at huge source activities and thicknesses, which are practically not used.",author = "V.
